PROJECT OVERVIEW
The Operetta CLSTM (Perkin Elmer) high-content analysis system is equipped with one camera and eight LED light sources and several filters (see table1). The instrument features a spinning disk confocal mode and can work with 5x air objectives but also with a 20x or 40x water objective supported with a water immersion kit, which is especially suited for imaging of zebrafish larvae. Currently Harmony version 4.8 is used to operate the high-content imager and to perform automated image analysis. We use the Preci-Scan function to detect zebrafish larvae in the well of interest in order to minimize measurement time and data generation. The software can be trained to detect certain zebrafish phenotypes, which is essential to carry out automated compound screens.
